      B7.1 Bromo Acetal Resins

      B7.1.1 HypoGel® Bromo Acetal Resins
      HypoGel® is a hydrophilic gel type resin which combines high capacities with good solvent compatibility. Glycol spacers with n = 5 or 10 EO units separate the reactive sites from the polystyrene matrix and modify the hydrophobic properties of the polystyrene backbone. Due to the spacer effect the functional group shows a higher mobility which results in higher kinetic rates and high resolution NMR spectra from immobilized compounds on the bead. The resin is compatible to a wide range of solvents and in contrast to polystyrene even polar solvents like methanol or ethanol can be used.
      These resins show high loading and a hydrophilicity between polystyrene and TentaGel®
      Reaction with primary amines generates immobilized N-substituted glycinal acetals suitable for the synthesis of heterocycles derived from 1-acyl-3-oxopiperazine.

      B7.1.2 TentaGel® Bromo Acetal Resins
      TentaGel® resins are grafted copolymers consisting of a low crosslinked polystyrene matrix on which polyethylene glycol (PEG or POE) is grafted. The PEG spacer is attached to the matrix via an ethyl ether group which increases stability towards acid treatment and minimizes PEG-leaching. As PEG is a "chameleon type" polymer with hydrophobic and hydrophilic properties, the graft copolymer shows modified physico chemical properties which are highly dominated by the PEG moiety (and no longer by the polystyrene matrix). These graft copolymers are pressure stable and can be used in batch processes as well as under continuous flow conditions. The PEG spacer is in the range of MW 3000 Da.
      Reaction with primary amines generates immobilized N-substituted glycinal acetals suitable for the synthesis of heterocycles derived from 1-acyl-3-oxopiperazine.

      HypoGel® is a hydrophilic gel type resin which combines high capacities with good solvent compatibility. Glycol spacers with n = 5 or 10 EO units separate the reactive sites from the polystyrene matrix and modify the hydrophobic properties of the polystyrene backbone. Due to the spacer effect the functional group shows a higher mobility which results in higher kinetic rates and high resolution NMR spectra from immobilized compounds on the bead. The resin is compatible to a wide range of solvents and in contrast to polystyrene even polar solvents like methanol or ethanol can be used.

      The Wang support is the most widely used resin for attaching carboxylic acids for further functionalisation. Cleavage conditions for the ester anchor involve concentrated TFA or TFA/CH?CI? mixtures to provide carboxylic acids or cyclization-cleavage under acidic or basic conditions. Release of an amide function by treatment with an amine in the presence of a Lewis acid was also reported. Wang resin was used to add vinyl and iodobenzoic acids via carbodiimide coupling and the products subjected to Heck conditions with alkenes or with alkynes or to Suzuki coupling with boron reagents. The Ugi multicomponent condensation reaction was conducted on Wang resin with the carboxylic acid, amine or isonitrile as the anchored functional group. On a Wang resin primary, secondary, and tertiary alcohols have been converted to 4-methoxy-benzyl ethers. As well as anchoring carboxylic acids to this resin, phenols were attached under Mitsunobu conditions, subjected to further transformations and cleaved under acidic conditions.
      Solid-phase lactam syntheses were also achieved by reaction of the resin-bound isonitrile with an amine and an w-keto-acid as well as other synthetic transformations such as the Biginelli reaction and construction of heterocycles such as quinolones, imidazoles, hydantoins, and 1,4-benzodiazepine-2,5-diones.

      TOPPA - A Novel Resin for Synthesis of Protected Peptide Amides
      This resin type is based on a TentaGel or polystyrene matrix and combines the very acid sensitive trityl handle with the acid sensitive Rink amide handle. The peptide synthesis can be performed by standard conditions. Treatment of the TOPPA resin with diluted AcOH, TFE or mixtures of both in dichloromethane cleaves the trityl ester bond and releases the total protected peptide amide (TOPPA). The C-terminus of the protected peptide amide is still masked by the Rink handle.

(containing 8% triisopropylsilane), AcOH/CF?OH/CH?CI?, 0.5% TFA or hexafluoroisopropanol. Recently, this resin has been used in cyclization reactions under Heck reaction conditions, the solid phase synthesis of b-peptides via the Arndt-Eistert homologation of Fmoc-protected amino acid diazoketones and in Mannich reactions of alkynes, secondary amines and aldehydes in the presence of a copper (I) salt affording the corresponding aminomethylalkynyl adducts.
      This resin can also be used in Fmoc peptide synthesis. Carboxylic acids can be released under very mild acidic conditions by treatment with AcOH/TFE/DCM or HFIP in DCM. Completely protected peptides are generated under this condition.

      HypoGel® is a hydrophilic gel type resin which combines high capacities with good solvent compatibility. Glycol spacers with n = 5 or 10 EO units separate the reactive sites from the polystyrene matrix and modify the hydrophobic properties of the polystyrene backbone. The hydrophilicity is between polystyrene and TentaGel® . Due to the spacer effect the functional group shows a higher mobility which results in higher kinetic rates and high resolution NMR spectra from immobilized compounds on the bead. The resin is compatible to a wide range of solvents and in contrast to polystyrene even polar solvents like methanol or ethanol can be used.

      B7.7.1 Polar PEG Spacer Resins with Base Labile Linkers
      B7.7.1.1 4-(Hydroxymethyl) Benzoic Acid (HMBA) on HypoGel® Resins
      HypoGel® is a hydrophilic gel type resin which combines high capacities with good solvent compatibility. Glycol spacers with n = 5 or 10 EO units separate the reactive sites from the polystyrene matrix and modify the hydrophobic properties of the polystyrene backbone. Due to the spacer effect the functional group shows a higher mobility which results in higher kinetic rates and high resolution NMR spectra from immobilized compounds on the bead. The resin is compatible to a wide range of solvents and in contrast to polystyrene even polar solvents like methanol or ethanol can be used.
      These resins show high loading and a hydrophilicity between polystyrene and TentaGel®
      This resin is derived from HypoGel® amine and the base labile 4-(hydroxymethyl)benzoic acid linker. It is a versatile support for the immobilization of carboxylic acids. The resulting ester bond is stable to strong acids but is cleaved by nucleophiles like amines, hydrazine or alkoxides.
